A major limitation of Polymerase Chain Reaction (PCR)-based research and diagnostic and forensic tests is the inhibition of DNA polymerases by many substances like as anticoagulants (e.g., heparin, EDTA, sodium citrate) and heme group, proteins, polysaccharides, cell debris, and charged inorganic compounds, etc. Therefore, DNA is usually extracted from specimen according to phenol-chloroform method or by an alternative ‘silica-membrane filter’ method. However, this is time consuming and labor intensive and there is also a risk of introducing contamination during the preparation.
